血管内皮因子-C_短发夹RNA靶向微泡联合超声辐照对裸鼠乳腺癌的抑制效应 被引量:1

摘要 目的探讨血管内皮生长因子-C_短发夹RNA(VEGF-C_shRNA)靶向微泡联合超声辐照对乳腺肿瘤生长的抑制作用。方法对32只雌性BALB/c裸鼠原位接种MCF-7人乳腺癌肿瘤组织。待移植瘤成瘤后,随机分为4组,G1组为空白对照,G2组采用VEGF-C_shRNA靶向微泡联合辐照,G3组为空白微泡+辐照,G4组为VEGF-C_shRNA静脉治疗。VEGF-C_shRNA靶向微泡治疗中,采用眼球后静脉注射150μg/kg两次,每次注射微泡后采用超声定位肿瘤后对微泡进行多次辐照。观察各组乳腺癌生长的情况。结果 G2组肿瘤增长幅度明显小于其他组,尤其是在治疗后第3、7、17天其平均增殖率与G1组差异有统计学意义(P均<0.05),G2组与G3组比较,第3、7、10、13、17及20天的差异均有统计学意义(P均<0.05),G2组与G4组第7、13天以及17天的差异有统计学意义(P<0.05)。结论 VEGF-C_shRNA靶向微泡联合辐照可以抑制裸鼠乳腺癌的生长。 Objective To assess the inhibitory effect of breast cancer for targeting vascular endothelial growth factor-C_short hairpin RNA(VEGF-C_shRNA) microbubble ultrasound with irradiation.Methods Thirty-two female BALB/c nude mice inoculated MCF-7human breast tumor tissue were randomly divided into 4groups : Control group(G1group),targeting VEGF-C_shRNA microbubble with irradiation(G2group),blank microbubble with irradiation(G3group) and VEGF-C_shRNA intravenous therapy(G4group).VEGF-C_shRNA of 150 μ g/kg was injected twice for targeted microbubble ultrasound treatment,with irradiation targeted for microbubble many times,then the growth of breast cancer was observed.Results The tumor growth degree of breast tumors of G2group was significantly less than other groups,especially on the 3rd,7th and 17th day,the mean proliferation rate was significantly different from that in G1group(all P 0.05).Compared with G3group,the mean proliferation rate of G2group was significantly different on the 3rd,7th,10th,13th,17th and 20th day(all P〈0.05).Compared with G4group,the mean proliferation rate of G2group was obviously different on the 7th,13th and 17th day(all P〈0.05).Conclusion VEGF-C_shRNA microbubble ultrasound with irradiation can inhibit the growth of breast cancer in nude mice.
