An LH radioreceptor assay (RRA) which allows direct measurement of receptor bindable bioactive serum LH in a variety of species was developed and validated. Using sheep luteal membrane as the receptor preparation source and 125 I labelled hCG as the tracer radioligand, the standard curve was obtained using high purified hCG. The assay is simple and can be completed within 2 hours. The sensitivity of the assay was 0.3 ng LH/per tube correspondent to hCG activity. The intra and interassay variations were (3.41±3.14)% and (8.70±7.93)%, respectively. Both the dose response curve of pituitary extract from bovine, swine, rabbit and the serum from bovine, ovine, swine, rabbit, rat, hamster showed parallelism to the hCG standard curve indicating that the RRA can be used to monitor the kinetic change of serum LH in a variety of species. The recoverable rates of 3, 10 and 30 ng hCG added to the diluted ram serum were (100.59±5.55)%, (99.55±9.86)% and (104.28±7.45)%, respectively, which demonstrated the veracity of the RRA results. The level of LH measured correlated with the physiological status of the animals. The bioactive LH level in pituitary extract was by far higher than that in circulation.
