白藜芦醇对脑缺血小鼠BDNF及NT-3表达的影响 被引量:10

摘要 目的探讨白藜芦醇(Resveratrol,Res)预处理对局灶性脑缺血小鼠的神经保护作用。方法将72只SPF级♂昆明小鼠随机分为假手术组、模型组、溶剂治疗组、白藜芦醇预治疗组,每组各18只。线栓法制作小鼠大脑中动脉永久性脑缺血模型(permanent middle cerebral artery occlusion,pM-CAO),免疫组织化学法检测脑组织BDNF及NT-3蛋白的表达,RT-PCR技术检测缺血脑组织中BDNF及NT-3的基因水平,2,3,5-氯化三苯基四氮唑(TTC)染色检测脑梗死体积的变化,以Zea Longa评分法进行神经功能学评分。结果与模型组及溶剂治疗组相比,白藜芦醇预治疗组梗死区周围BDNF及NT-3的表达水平均明显升高(P<0.01),脑梗死体积明显缩小(P<0.01),且神经功能学评分明显改善(P<0.01)。结论白藜芦醇可增加脑缺血后BDNF及NT-3的表达,减少脑梗死体积,改善神经功能缺损。 Aim To investigate the neuroprotective effects of Resveratrol pretreatment after cerebral ischemia in mice.Methods SPF male Kunming mice(n=72) were randomly divided into 4 groups:sham-operated group,ischemia group,ischemia+vehicle group and ischemia+Resveratrol group(Each group of 18 mice).Immunohistological and RT-PCR assessments were performed to evaluate the effects.The pathological changes of brain tissue and infarct volume were observed by 2,3,5-triphenytetrazolium chloride(TTC) staining,and the score of neurological function was evaluated by Zea-Longa test.Results Compared with the ischemia and ischemia+vehicle groups,the expression of BDNF and NT-3 obviously increased(P0.01) in Resveratrol group,but the infarct volume and the neurological function scores were decreased significantly in Resveratrol group(P0.01).Conclusion Resveratrol can improve the neurological function through decreasing the infarct volume and increasing the expression of BDNF and NT-3 after cerebral ischemia in mice.
