摘要
目的探讨原花青素对Ⅱ型糖尿病大鼠心肌组织损伤保护作用及其机制。方法 SD大鼠随机分为5组:对照组、糖尿病模型组、原花青素低、中、高剂量组。后4组采用小剂量链尿佐菌素30 mg/kg,制备Ⅱ型糖尿病大鼠模型。原花青素低、中、高剂量组分别按50、100、200 mg/kg灌胃12周。检测血糖及血脂,心功能状态;免疫组织化学法和western blot法测定心肌组织核转录因子(NF-κB)、肿瘤坏死因子(TNF-α)、黏附因子-1(ICAM-1)和单核趋化蛋白-1(MCP-1)蛋白表达。结果与对照组比较,糖尿病模型组大鼠血糖及血脂明显增高(P<0.01),心肌组织中NF-κB、TNF-α、ICAM-1、MCP-1蛋白表达明显增加(P<0.05);与糖尿病模型组比较,原花青素中剂量组血糖、TG、TC、LDL明显降低(P<0.05),HDL明显提高(P<0.05);LVEDP[(4.65±0.96)mmHg]明显降低(P<0.05),LVSP[(125.58±5.65)mmHg]明显升高(P<0.05),NF-κB、TNF-α、ICAM-1、MCP-1蛋白表达分别为11.24、0.19、4.57、3.15明显减少(P<0.05)。结论原花青素对Ⅱ型糖尿病大鼠心肌有保护作用,其机制可能与降低血糖、血脂、抑制炎性反应有关。
Objective To explore protective effect of procyanidins on myocardial tissue damage in the rats with type Ⅱdiabetes and its mechanism.Methods SD rats were randomly divided into control group,diabetes model group,low,moderate and high dose procyanidins groups.Low dosage streptozocin(STZ,30 mg/kg) was used to induce type Ⅱ diabetes model of rat.Procyanidins at doses of 50,100,and 200 mg/kg was administered with stomach gavage for 12 weeks.Blood sugar and fat and heart function of the rats were detected.Immuno-histochemistry methods were used to determine levels of nuclear factor-κB(NF-κB),tumor necrosis factor-alpha(TNF-α),intercelluar adhesion molecualr-1(TCAM-1),monocyte chemoatracttant protein-1(MCP-1) in myocrdial tissue of the rats.Results Compared with the controls,blood sugar and blood fat increased significantly in diabetic rats(P〈0.01).The levels of NF-k B,TNF-a,ICAM-1,and MCP-1 in myocardial tissue of diabetic rats increased significantly(P〈0.05).Compared with the diabetes model,blood sugar,triglyceride,total cholesteral,and low-density lioprotin declined significantly(P〈0.05),while high-density lipoprotein increased significantly(P〈0.05) in the rats with porcyanidins treatment.In addition,the left ventricular end-diastolic pressure was declined significantly(P〈0.05) and left indoor pressure rise rate,±dp/dtmax were elevated(P 0.05) for the rats treated with procyanidins.The levels of NF-κB,TNF-α,ICAM-1,and MCP-1 in moderate dose procyanidins treatment group were 11.24,0.19,4.57,and 3.15,with significant reductions(P〈0.05).Conclusion Procyanidins have protective effects on myocardial tissue in rats with type Ⅱ diabetes and the effects may related to the reductions of blood sugar and lipids and the inhihition of inflummatory reaction.
