摘要
目的:探讨归脾汤对小鼠镇静催眠及记忆巩固性障碍的影响。方法:将小鼠随机分为空白组、阳性组、归脾汤高剂量组、归脾汤中剂量组、归脾汤低剂量组。阳性组给予安定片5 mg.(kg.d)-1和安神补脑液800 mg.(kg.d)-1治疗,归脾汤高剂量用药为归脾汤2 000 mg.(kg.d)-1、归脾汤中剂量组用药为归脾汤1 000 mg.(kg.d)-1、归脾汤低剂量组用药为归脾汤500 mg.(kg.d)-1。测定5 min内小鼠的活动次数和站立次数,记录各组小鼠的睡眠潜伏期、睡眠持续时间和测定小鼠脑内单胺类神经递质含量;小鼠皮下注射亚硝酸钠(NaNO2)120 mg.kg-1,制得记忆巩固障碍小鼠模型,记录小鼠第一次跳下跳台的时间(即触电潜伏期)及小鼠10 min内跳下的次数(即错误次数)作为记忆指标。结果:小鼠的活动次数和站立次数明显减少(P<0.05),能延长戊巴比妥钠所致小鼠的睡眠时间(P<0.05或0.01),明显地增加小鼠大脑组织中5-羟色胺(5-HT),多巴胺(DA),去甲肾上腺素(NA)和5-羟吲哚乙酸(5-HIAA)的含量(P<0.05或0.01),触电潜伏期有一定的延长,均可减少小鼠10 min内的错误次数(P<0.05)。结论:归脾汤有一定的镇静催眠和改善学习记忆的作用。
Objective:To study the effect of Guipi Decoction on the sedative-hypnotic and memory consolidation obstacles of mice.Methods:The mice were randomly divided into the control group,positive group,high dose group of Guipi Decoction,medial dose group of Guipi Decoction and low-dose group of Guipi Decoction.The positive group was given diazepam 5 mg·(kg·d)-1 and Anshenbunao Syrup 800 mg·(kg·d)-1 for treatment.The high dose group was given 2000 mg·(kg·d)-1 of Guipi Decoction;the medial dose group was given 1000 mg·(kg·d)-1 of Guipi Decoction;the low dose group was given 500 mg·(kg·d)-1 of Guipi Decoction.The number of activities and the standing number of the mice within 5 min were determined.The sleep latency,sleep duration and the content of mouse brain monoamine neurotransmitter of the mice in each group were recorded and determined.The mice were given subcutaneous injection of sodium nitrite(NaNO2 120 mg·kg-1) to build mice models with memory consolidation obstacles.The time of the mice jumping off the platform(ie,electric shock latency) and the number of jumping within10 min(ie,the number of errors) were recorded as the memory index.Results:The number of activities and standing up of the mice decreased(P0.05).It could extend the mouse′s sleeping-time caused by napental(P0.05 or 0.01).It also could significantly increase 5-HT,DA,NA and 5-HIAA levels in the brain tissue of the mice(P0.05 or 0.01).Beside,it also could extend a certain incubation period of electric shock,and reduce the number of errors of the mice within 10 min(P0.05).Conclusion:Guipi Decoction has obviously sedative-hypnotic effect and the function of improving the learning abilities and memory.
