雾化吸入和鼻内滴注嗜热吸水链霉菌诱导豚鼠大棚肺模型的比较 被引量:2

Comparison of guinea pig green-house lung induced by ultrasonic atomizing inhalation and intranasal instillation of streptomyces thermohydroscopicus

摘要 目的探讨雾化吸入嗜热吸水链霉菌抗原制剂能否诱导豚鼠大棚肺模型及比较雾化法和鼻滴法二者的差异。方法成年豚鼠40只,体重300~400g,雌性,随机分为模型组(雾化组A1、鼻滴组B1)和对照组(雾化组A2、鼻滴组B2),每组20只。A1组:嗜热吸水链霉菌抗原制剂5mL日1次雾化吸入,15min/次,连续12周;B1组:0.3mL抗原制剂鼻内滴注,连续3次/周,连续12周;A2组:0.9%无菌生理盐水5mL日1次雾化,15min/次,连续12周;B2组:0.9%无菌生理盐水0.3mL鼻滴,连续3次/周,连续12周。结果①影像学改变:A1组:第1周末未见异常;第2周末,可见双肺弥漫性磨玻璃影;第3周末,可见双肺微小结节影;第12周末,可见明显的纤维条索影。B1组:第1周末,可见双肺磨玻璃样改变;第2周末,可见双肺多发斑片状影;第3周末,可见双肺散在小结节影;第12周末,可见纤维条索影。对照(A2、B2)组无异常。②病理改变:A1组:第1周末未见异常;第2周末,可见肺泡炎改变;第3周末,可见肉芽肿改变;第12周末,可见纤维化改变。B1组:第1周末,可见肺泡炎改变;第2周末,可见渗出性改变;第3周末,可见肉芽肿改变;第12周末,可见纤维化改变。对照(A2、B2)组无异常。③肺组织研磨后培养,A1、B1组均可见嗜热吸水链霉菌生长。结论雾化吸入嗜热吸水链霉菌抗原制剂可诱导豚鼠大棚肺模型;雾化法致豚鼠大棚肺肺泡炎改变迟于鼻滴法1周出现。 【Objective】 To investigate whether ultrasonic atomizing inhalation of the streptomyces thermohydroscopicus can cause the green-house lung and the differences of ultrasonic atomizing inhalation and intranasal instillation.【Methods】 Forty guinea pigs(female,weighed 300 to 400 g) were randomly divided into four groups(10 guinea pigs per group): Group A1: ultrasonic atomizing inhaled streptomyces thermohydroscopicus antigen 5 mL per day,12 consecutive weeks;Group B1: intranasal instilled streptomyces thermohydroscopicus antigen 0.3 mL during 3 consecutive days per week,12 consecutive weeks;Group A2: ultrasonic atomizing inhaled 5 mL 0.9% saline per day,12 consecutive weeks;Group B2: intranasal instilled 0.3 mL 0.9% saline during 3 consecutive days per week,12 consecutive weeks.【Results】 ①Iconography changed: Group A1: the 1st week,the CT was normal;the 2nd week,ground glass opacity changed in both lung;the 3rd week,diffused micro tuber in both lung;the 12th week,fibrosis changed in both lung.Group B1: the 1st week,ground glass opacity changed in both lungs;the 3rd week,diffused micro tuber in both lungs;the 90th day,fibrosis changed in both lungs.②Pathology changed: Group A1: the 1st week,was normal;the 2nd week,it appeared alveolitis;the 3rd week,granuloma changed;the 12th week,the number of fibroblasts increased.Group B1: the 1st week,it appeared alveolitis;the 3rd week,granuloma changed;the 12th week,it appeared fibrosis.③Lung grinded and cultured could see streptomyces thermohydroscopicus in Group A1,B1.【Conclusions】 Ultrasonic atomizing inhalation of the streptomyces thermohydroscopicus can cause the model of green-house lung in the guinea pigs;The change of alveolitis is one week later by ultrasonic atomizing inhaled than intranasal instilled.
