雌激素受体与雌激素非靶器官肿瘤的研究进展

Research advances in the estrogen receptors in estrogen non-target organ tumors

目的:总结经典雌激素核受体ERα和ERβ在非靶器官肿瘤中的研究进展。方法:以"雌激素受体、非靶器官和肿瘤"等为关键词,检索2005-01-2011-06PubMed以及CNKI期刊全文数据库相关文献,共检索到相关文献153篇。排除标准:靶器官和非肿瘤。纳入标准:1)雌激素受体与肿瘤相关性及其机制;2)雌激素非靶器官肿瘤及基础与临床研究。根据纳入标准,符合分析的文献61篇。结果:雌激素不仅对人体生长、发育和器官功能维持等起重要的生理作用,而且与众多肿瘤的进展密切相关。雌激素主要通过雌激素受体(ER)发挥作用。ER在雌激素靶器官肿瘤过表达,雌激素对ER阳性靶器官肿瘤有显著的促生长作用,ER抑制剂在部分雌激素靶器官肿瘤的临床治疗中占据重要地位。近年来,雌激素与非靶器官肿瘤的相关性逐渐受到关注,ER在雌激素非靶器官肿瘤中的广泛表达提示,ER与非靶器官肿瘤的发生、发展也存在密切关系。结论:对ER与非靶器官肿瘤的深入研究可揭示雌激素及其受体在非靶器官肿瘤发生、发展中的作用及机制,并有可能发现肿瘤治疗的新靶点,为雌激素非靶器官肿瘤的防治提供新途径。

OBJECTIVE： To sum up current and potential clinical relevance about ER α and ER β in non-target organ tumors.METHODS： The retrieval system of PubMed and CNKI Academic Journal were used to search documents about estrogen receptors from 2005-01 to 2011-06,with ＂estrogen receptor,non-target organ,tumor＂ as the key words Standards of getting in：estrogen receptor associated with tumors and its mechanism,estrogen non-target organ tumors and basic/clinical research.Standards of getting out： target organ,non-tumor.According to the above standards,61 of the articles were chosen and summarized for study.RESULTS： Estrogen,via estrogen receptor（ER）,plays an important role in the physiological functions of growth,development,and organ function maintenance of human body,at the same time contribute to the progression of a great number of tumors.The over expression of ER was demonstrated in target organ tumors.And estrogen promotes the growth of ER-positive target organ tumors significantly.ER inhibitors occupy an important position in the clinical treatment of tumors in some estrogen target organ.However,the correlation between estrogen and tumors of non-estrogen-target organ has been drawing more and more attention in recent years.The extensively expression of ER in the tumors of non-estrogen-target organ suggested they may be in close relation in tumor development.Recently,more progression in the researches of the classic estrogen receptors,ER alpha and beta,had been reported.CONCLUSION： Further research can reveal the role and mechanism of estrogen and its receptor in the development of tumors in non-target organ,uncover new effective targets in cancer treatment,and provide new approaches to the prevention and treatment of tumors in non-estrogen-target organ.
