奥曲肽玻璃体腔注射对视网膜新生血管的抑制作用及其机制探讨 被引量:1

摘要 目的:探讨奥曲肽对视网膜新生血管形成的抑制作用及其对视网膜上VEGF和IGF-1表达的影响。方法:将健康的鼠龄7d的昆明小鼠24只随机分为3组,每组8只,正常组(A组)于正常空气环境中饲养,实验组(B组)和对照组(C组)置于氧箱中饲养。鼠龄12天时,B组玻璃体腔内注射奥曲肽3μl,A组和C组玻璃体腔内注射等量生理盐水,给药后第7天,将小鼠处死,摘除眼球制作组织学标本,视网膜铺片光镜下观察视网膜血管形态;HE染色计数突破内界膜的内皮细胞核平均数;免疫组化定量分析VEGF和IGF-1在视网膜上的表达。统计学分析统计结果以均数士标准差表示,应用SPSS13.0软件进行统计分析,采用方差分析,组间两样本比较采用LSD-t检验,P<0.05认为差异有统计学意义。结果:A、B、C组HE染色突破视网膜内界膜的血管内皮细胞核平均数呈增加趋势,各组间差异显著(P<0.05),A、B、C组视网膜上VEGF和IGF-1的阳性表达有增强趋势,各组间差异显著(P<0.05),视网膜上VEGF与IGF-1的表达呈显著正相关性(P<0.05)。结论:奥曲肽能有效抑制视网膜新生血管的形成,同时降低VEGF和IGF-1在视网膜上的表达水平。
