不同预后小肠间质瘤的比较蛋白质组学分析

摘要 目的:分析不同预后小肠间质瘤病人组织蛋白质质点,寻找判断小肠间质瘤预后的候选蛋白质。方法:预后不良组和预后良好组小肠间质瘤组织各5例,经双向凝胶电泳分离总蛋白质,运用图像分析软件分析,对差异蛋白质点采用基质辅助激光解析电离飞行时间质谱仪鉴定部分差异蛋白质点。结果:比较蛋白质组学共鉴定出蛋白质28个(预后不良组中9个蛋白质表达上调,19个蛋白质下调)。预后不良组小肠间质瘤组织中微管不稳定蛋白(stathmin)、血清结合珠蛋白(haptoglobin 2,HP-2)和微管蛋白表达明显升高。结论:不同预后小肠间质瘤组织的蛋白质谱存在差异,stathmin和HP-2及微管蛋白可能成为判断小肠间质瘤病人预后的3种因子,为今后小肠间质瘤病人的临床预后判断提供依据。 Objective To analyze the proteomic components of the tissues from the patients with small bowel stromal tumors(SBSTs) with different prognosis,and to search the candidate prognostic biomarkers for the SBSTs.Methods Proteins from tissues of 5 cases with SBSTs with good prognosis and 5 cases with SBSTs with poor prognosis were separated by two-dimensional gel electrophoresis(2-DE),analyzed by image analysis software and identified by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry(MALDI-TOF-MS).Results A total of unique 28 different proteins were identified by comparative proteomic analysis.Among them,9 proteins are obviously up-regulated and 19 proteins down-regulated in the poor prognosis group.MALDI-TOF-MS revealed that the expression of stathmin,HP-2 and tubulin were significantly up-regulated in the poor prognosis group.Conclusions Protein spectra are different in tissues from SBSTs patients with different prognosis.Stathmin,HP-2 and tubulin may be three potential prognostic markers for SBSTs.
