液相芯片技术检测卵泡液中细胞因子水平的可行性研究 被引量:3

摘要 目的:初步评价液相芯片技术用于检测控制性超促排卵患者取卵时卵泡液中36种细胞因子水平的可行性。方法:采用液相芯片技术检测81名不孕症患者单一卵泡卵泡液中36种细胞因子水平,通过检测结果初步评价其检测卵泡液中细胞因子水平的可行性。结果:液相芯片技术应用于检测卵泡液中白介素(IL)-1β、IL-4、IL-5、IL-10、瘦素(Leptin)、巨噬细胞炎性蛋白(MIP)-1β、肿瘤坏死因子(TNF)-β、血管内皮生长因子(VEGF)水平时,检测的准确度、精密度、敏感性均较高,结果可信。结论:液相芯片技术可用于检测卵泡液中细胞因子水平,但仍须进一步完善检测系统的评价及提高检验灵敏度,使可检测的卵泡液中目的细胞因子范围扩大,以提高其在卵泡液细胞因子水平检测中的应用价值。 Objective:To preliminarily evaluate the feasibility of applying bead-based multiplex sandwich immunoassay(Luminex) to assay 36 cytokines in the follicular fluid(FF) obtained during oocytes retrieval from patients underwent controlled ovarian hyperstimulation(COH).Methods:The levels of 36 cytokines in the follicular fluid obtained from a single follicle of 81 infertility patients were measured by Luminex.Then the feasibility of applying Luminex to measure cytokines in the FF was preliminarily evaluated.Results:When using Luminex to assay the levels of interleukin(IL)-1β,IL-4,IL-5,IL-10,leptin,macrophage inflammatory protein(MIP)-1β,tumor necrosis factor(TNF)-β,vascular endothelial growth factor(VEGF),the results were credible because the measurement accuracy,precision and sensitivity were high.Conclusion:Luminex can be used to assay levels of cytokines in FF.But to broaden the test scope of cytokines aimed at in the follicular fluid,it needs to perfect the evaluation of the testing system and further improve the measurement sensitivity.
