电针对Cardiotoxin肌肉损伤小鼠MyoD表达的影响 被引量:6

摘要 目的探讨电针对肌肉损伤小鼠MyoD表达的影响,为针灸在临床上治疗各类肌肉损伤引起的肌肉再生不足提供实验依据。方法将蛇毒细胞毒素(CTX,Cardiotoxin)胫骨前肌注射造成的肌肉损伤小鼠随机分为模型组和CTX电针组。电针治疗7d后取胫骨前肌制作冰冻切片,HE染色观察损伤肌肉的形态学变化、免疫组化染色检测MyoD的表达。结果 HE染色显示2组均可见部分肌纤维形态破坏,肌细胞间质中炎症细胞浸润,而电针组炎症细胞浸润较模型组明显增多。免疫组化显示模型组可见MyoD表达,电针组MyoD阳性细胞面积百分比较模型组明显增高(P<0.01)。结论电针能够促进肌肉损伤后其他细胞向成肌细胞的转化,对肌肉再生有增强作用。 OBJECTIVE To study the effect of electroacupuncture(EA) on the expression of MyoD in mice with muscle injury,and to provide experimental evidence for treating muscle regeneration deficiency in clinics.METHODS Mice with musle injury induced by Cardiotoxin injected in tibialis anterior were randomly assigned into 2 groups:model group and EA group.After 7 days EA therapy,tibialis anterior muscles were collected for preparation of frozen sections.HE staining was performed for examination of the pathological changes in the injured muscle,and immunohistochemistry staining were used to detect protein expressions of MyoD.RESULTS HE staining showed that the morphous of myofiber was destroyed partly,and inflammatory cell infiltration was observed in two groups.But the EA group showed a higher inflammatory cell numbers than that in model group.The expression of MyoD was observed in two groups,but the expression in EA group was obviously higher than that in model group(P0.01).CONCLUSION EA could promote the transformation of other cells to myogenic cell,thus playing a positive role in the muscle regeneration.
