CD_(44)和E-cadherin表达与肾癌细胞增殖潜能的研究 被引量:3

The significance of CD_(44) and E-cadherin expressions as related to the proliferative potential of human renal carcinoma cells

摘要 目的 探讨CD4 4和E cadherin分子表达与人肾癌细胞增殖潜能的关系。 方法 建立人肾癌GRC 1和RLC 310细胞的克隆性亚细胞株 ,检测其克隆形成率 ,应用免疫组织化学法检测CD4 4和E cadherin分子表达 ,同时用流式细胞术分析CD4 4分子表达。 结果 RLC 310细胞的三个亚株RLC 310A ,RLC 310B和RLC 310C在软琼脂上的集落形成率分别为 48% ,5 0 % ,30 % ,CD4 4阳性表达率为 10 .6 % ,15 .6 %和 6 5 .0 % ;GRC 1细胞的三个亚株GRC 1A ,GRC 1B和GRC 1C在软琼脂上均不形成集落 ,CD4 4阳性表达率分别为 5 1.3% ,6 4.8%和 83.5 %。两株细胞的不同亚株E cad herin表达均为强阳性。 结论 肾癌细胞株均表达CD4 4和E cadherin ,CD4 4表达下降与肾癌细胞的增殖潜能增加有关 ,E cadherin与肾癌细胞的增殖能力变化无明显关系。 Objective To investigate CD 44 and E cadherin expressions and their correlation with the proliferative potential of human renal cell carcinoma cells. Methods The subclonal cell lines of human renal cell carcinoma GRC 1 and RLC 310 were established and their clongenic efficiency was counted.Immunohistochemical staining for CD 44 and E cadherin in the subpopulation cell lines was carried out and CD 44 expression was also studied by flow cytometry(FCM). Results The colony efficiency of the subclonal cell lines RLC 310A,RLC 310B and RLC 310C in soft agar was detected in 48%、50% and 30% respectively.CD 44 staining was positive(+)in RLC 310A,RLC 310B and highly positive (+++) in RLC 310C and were counted in 10.6%,15.6% and 65.0% respectively.No colony formation in soft agar was observed in subclonal cell lines of GRC 1.CD 44 staining was highly positive in GRC 1A,GRC 1B,GRC 1C and was counted in 51.3%,64.8%and 83.5% respectively.E cadherin staining was highly positive in all the subclonal cell lines. Conclusions CD 44 and E cadherin expressions were positive in human renal cell carcinoma cell lines.The decrease of CD 44 expression was correlated with increasing of proliferating potential of renal cell carcinoma cells whereas E cadherin expression did not.
