摘要
目的 :探讨肾康注射液 (SKI)阻抑肾小球硬化 (GS)、防治慢性肾功能衰竭 (CRF)的部分作用机理 ,为SKI临床应用提供依据。方法 :采用放射免疫检测技术 ,观察SKI对体外培养人肾小球系膜细胞 (GMC)自分泌内皮素 1(ET 1)、一氧化氮 (NO)水平的影响。结果 :SKI及其含药血清明显抑制GMC自分泌ET 1(P <0 .0 1、P <0 .0 5)、促进其自分泌NO(P <0 .0 5) ,有量效依赖性 ;与肝素合用 ,协同抑制GMC自分泌ET 1水平 (P <0 .0 5)。结论 :SKI对GMC自分泌ET 1、NO的调节作用是其阻抑GS、防治CRF的重要机制之一。SKI对ET 1的抑制效应、对NO的促进作用是其活血益气功效的具体形式之一。
Objective:To approach the mechanism of SKI on prohibiting Glomerulosclerosis(GS) so that treatment on chronic renal failure(CRF) with SKI furnishs fresh evidence in the level of cytobiology.Method:Using serumpharmacological test of Chinese medicinal herbs,we observed that how did SKI stimulates endothelin 1(ET 1) and nitric oxide(NO) secretion in cultured human glomerular mesangial cell(GMC).Result:SKI and SKI serum restrained significantly level of ET 1 and promoted NO secretion in GMC(P<0.05),showing dependence on concentration.Working together with herparin,effect of SKI could been strengthened.Conclusion:Modulating production of NO,ET 1 is one of the mechanism of SKI on prohibiting GS,and the important link of SKI on preventing and treating CRF.
