摘要
目的 :建立荧光分光光度法测定人血浆中左氧氟沙星浓度。方法 :以三氯乙酸沉淀蛋白 ,分离后加醋酸 -醋酸钠缓冲液 (p H4.0 )测荧光强度 ,激发波长 2 94nm,发射波长 496 nm。结果 :左氧氟沙星浓度在 0 .1~ 5 .0 μg· m l- 1范围内有良好的线性关系 ,回归方程为 Y=2 4.916 X- 1.2 97,r=0 .9994。平均回收率为 98.5 2 % ,RSD为 3.12 %。结论 :该方法简便。
OBJECTIVE:A spectrofluorometry was used for determination of levofloxcin concentration in human plasma.METHODS:Human plasma protein was precipitated with trichloroacetic acid and fluoroscent intensity of levofloxacin was measured in acetic acid sodium acetate buffer(pH4.0).Its λex and λem was 294nm and 496 nm ,respectively.RESULTS:Linear relationship was found in the range from 0.1 to 5.0 μg·ml 1 . The regression equation was Y=24.916X 1.297.The correlation coefficient(r) was 0.9994.The average recovery was 98.52%.The coefficient of variation was 3.12%.CONCLUSION:The method is simple,rapid and accurate.
