摘要
目的 探讨阿尔茨海默病 (AD)与淀粉样 β蛋白前体 (APP)基因的 16和 17外显子表达之间的关系。方法 应用相对定量的竞争RNA扩增聚合酶链反应 (RT PCR)技术 ,测定 5 2例AD患者、2 8例血管性痴呆 (VD)患者及 6 0名健康老年人的外周血单核细胞中APP基因 16和 17外显子的表达量。结果 AD患者、VD患者及健康老年人每微克cDNA的APP基因的 16和 17外显因子表达量分别为 (4 0 5± 0 2 7)amol,(2 73± 0 33)amol和 (2 5 9± 0 2 7)amol,其中AD患者表达量高于健康老年人(t=3 79,P <0 0 1)和VD患者 (t=4 13,P <0 0 1)。结论 AD患者的外周血单核细胞中可能存在APP基因的 16和 17外显子过度表达的特异性。
Objective To clarify the relation between the expression of β amyloid precursor protein gene (16 and 17 exons) and Alzheimer disease(AD) Methods The expression of β amyloid precursor protein gene in peripheral mononuclear blood cell (PMBC) was examined using quantitive competitive RT PCR methods in 52 AD cases, 28 vascular dementia (VD) patients and 60 older healthy controls Results In every μg cDNA,the expression of APP in PMBC were (4 05±0 27) amol, (2 73±0 33) amol and (2 59±0 27) amol respectively Obviously, the APP gene (16 and 17 exons) was overexpressed in AD compared with health controls( t =3 79, P <0 01)or VD cases( t =4 13, P <0 01) Conclusion The 16 and 17 exons overexpression of APP gene in PMBC might be specific in AD cases
