摘要
目的 探讨载脂蛋白E(apoE)基因与神经系统记忆功能之间的关系。方法 以单克隆抗体酶联免疫吸附测定法分别测定apoE4 、apoE7转基因鼠及对照组鼠 (每组各 12只 )的血清apoE含量。通过主动回避反应 (跳台 )试验对apoE4 、apoE7转基因鼠及对照组鼠的神经系统记忆功能进行检测。结果 人apoE基因不仅能在F1代仔鼠体内表达 ,且血清apoE含量 [apoE4 鼠 (1 72± 0 34 )mg/L ,apoE7鼠 (1 5 3± 0 2 4)mg/L]与对照组鼠 [(0 76± 0 0 7)mg/L]的差异均有显著性 (P <0 0 5 )。apoE4 转基因鼠在学习 [(2 1± 2 1)min]和短期记忆 [(1 2± 1 2 )min]方面所表现出的受电击时间总和均长于对照组鼠 [(0 4± 0 3)min ,(0 1± 0 1)min],P <0 0 5 ;而apoE7转基因鼠则只表现为短期记忆方面的错误次数 (2 2± 2 1)高于对照组鼠 (0 8± 1 6 ) ,P <0 0 5。结论 提示apoE基因的异常表达影响转基因鼠的大脑功能。
Objective The aim of this study was to investigate the relationship between the apolipoprotein E gene and memory function Methods The human apoE level was measured in the serum of apoE 4 and apoE 7 transgenic mice with ELISA methods The memory function of transgenic mice was determined on active avoidance response Results The human apoE gene could be expressed in the first generation of transgenic mice The apoE levels of transgenic mice (apoE 4 mice [(1 72±0 34)mg/L, apoE 7 mice(1 53±0 24)mg/L] were significant differed with control mice [(0 76±0 07)mg/L], P <0 05 The learning ability [(2 1±2 1)min] and short time memory [(1 2±1 2)min] were prominently increased in the apoE 4 transgenic mice compare with the controls [(0 4±0 3)min and(0 1±0 1)min], P <0 05 More mistake number of short time memory could be observed in apoE 7 transgenic mice [(2 2±2 1) times] compare with control mice [(0 8±1 6 )times], P <0 05 Conclusion The results showed that the abnormal express of apoE gene might result in great injury on the brain function in the transgenic mice brains
