P53mRNA及P53蛋白在肾上腺嗜铬细胞瘤中的表达及意义 被引量:2

摘要 目的 :研究抑癌基因 P5 3及其表达产物 P5 3蛋白在肾上腺嗜铬细胞瘤中的表达 ,比较 P5 3m RNA的表达与 P5 3蛋白的表达关系。方法 :应用免疫组织化学方法及核酸分子原位杂交方法检测 P5 3m RNA及 P5 3蛋白的表达情况。结果 :原位杂交显示 ,P5 3m RNA在肾上腺嗜铬细胞瘤中表达为 2 4.1% (7/ 2 9) ,在肾上腺恶性嗜铬细胞瘤中表达为 14.3% (1/ 7) ;免疫组织化学显示 ,P5 3蛋白在肾上腺嗜铬细胞瘤中表达为 79.3% (2 3/ 2 9) ,在肾上腺恶性嗜铬细胞瘤中表达为 71.4% (5 / 7)。结论 :在肾上腺嗜铬细胞瘤中 ,存在 P5 3基因突变 ,推测 P5 Purpose:To investigate the expression and mechanism of antioncogene P53 mRNA and its protein in human adrenal pheochromocytoma.Methods:In situ hybridizaiton (ISH) was employed to detect P53 mRNA in pheochromocytoma.P53 protein was examined by SP immunohistochemistry.Results:ISH showed the expression of P53 mRNA in 29 cases of pheochromocytomas was 24.1 % (7/29) and in 7 cases of malignant pheochomocytomas was 14.3 % (1/7).The expression of P53 protein in 29 cases of pheochromocytomas was 79.3 % (23/29) and in 7 cases of malignant pheochromocytomas was 71.4 % (5/7).Conclusions:The mutation of P53 gene might be very important in and responsible for pathogenesis of pheochromocytoma.
