丹红注射液预处理对大鼠心脏缺血再灌注损伤的保护作用 被引量:1

摘要 目的探讨丹红注射液预处理对大鼠心脏缺血再灌注损伤的保护作用。方法雄性SD大鼠24只。大鼠心脏离体后悬于Langendorff灌流装置。随机分为3组,每组8只:缺血组(ISCH组):大鼠心脏离体平衡50min,37℃缺血45min,K-H液复灌3h;低浓度丹红注射液预处理组(L-prec组):大鼠心脏平衡20min,以含低浓度丹红注射液(5mg/L)的灌注液灌注15min,洗脱15min,37℃缺血45min,K-H液复灌3h;高浓度丹红注射液预处理组(H-prec组):大鼠心脏平衡20min,以含高浓度丹红注射液(15mg/L)的灌注液灌注15min,洗脱15min,37℃缺血45min,K-H液复灌3h。平衡50min和复灌2h期间连续监测LVDP,LVDEP,±dp/dtmax。复灌60min时接取冠脉流出液,测定冠脉流出液中CK和LDH的含量。复灌结束,将心脏切片(1~2mm)并做TTC染色。结果机械功能:基础状态时3组LVDP,LVDEP,±dp/dtmax两两比较差异无统计学意义(P>0.05)。复灌期间L-prec组的LVDP,LVDEP,±dp/dtmax与ISCH组相比,差异无统计学意义。H-prec组的LVDP和±dp/dtmax与ISCH组相比,显著升高(P<0.05);LVDEP与ISCH组相比,显著降低(P<0.05)。冠脉生化指标及心梗面积:复灌60min时L-prec组和H-prec组的CK和LDH及心梗面积与ISCH组相比,显著性降低(P<0.05);复灌60min时L-prec组和H-prec组之间CK和LDH及心梗面积差异无统计学意义(P>0.05)。结论丹红注射液预处理能减轻心肌缺血再灌注损伤后CK和LDH的释放,缩小心梗面积;高浓度丹红注射液预处理能显著增强心脏缺血再灌注损伤后心脏的机械功能。 Objective This study investigated whether Danhong Injection could protect perfused hearts of rats against ischemia/reperfusion injury (IR) when it was administered before the adverse isehemic event. Methods Studies were conducted in buffer-perfused isolated rat hearts. This experiment consisted of baseline perfusion for 20 min, warm global ischemia for 40 min and 2 hours of reperfusion. Preconditioning of hearts in L- prec and H-prec group was induced by 15 min of low(5 mg/L) or high( 15 mg/L) concentration of Danhong In- jection followed by 15 min of wash out. Untreated hearts served as isehemic control (ISCH) group. Left ventric- ular mechanical function (LVDEP, LVDP, ±dp/dtmax) , cardiac enzyme aetivity(CK, LDH), and infarct size (IS) were measured as variables of ventricular function and cellular injury, respectively. Results Dan- hong Injection preconditioning attenuated the release of CK and LDH and reduced the IS of the heart during reperfusion, compared with ISCH group (P 〈 0. 05 ). There was no difference between L-prec and H-prec group with respect to myocardial enzyme activity and infarct size. Additionally, high concentration of Danhong Injec- tion improved mechanical function comparing with ISCH group and L-pree group ( P 〈 0. 05 ). Conclusion Danhong Injection administered before the ischemia provides protective effects against reperfusion injury in isola- ted rat hearts, high concentration of Danhong Injection improved mechanical function.
