摘要
目的研究新安鼻渊方对大鼠急性鼻窦炎的防治作用。方法 90只SD大鼠随机留取10只作为正常对照组,其余80只大鼠用于复制急性鼻窦炎模型,根据鼻窦炎症状评分标准,选取评分标准≥6分的大鼠60只随机分为模型组,新安鼻渊方高(15.2g/kg)、中(7.6g/kg)、低(3.8g/kg)剂量组,鼻渊舒组(3.5ml/kg),左氧氟沙星组(40mg/kg),每组10只。模型复制7d后开始灌胃给药,正常组、模型组以等容量生理盐水灌胃,给药时间为7d。观察大鼠鼻窦炎症状评分、体温、血液白细胞计数、中性粒细胞构成比、血中溶菌酶含量、鼻窦病理变化。结果模型组大鼠鼻窦炎症状评分、体温、白细胞计数、中性粒细胞构成比均显著高于正常对照组;左氧氟沙星组、鼻渊舒组、新安鼻渊方各剂量组大鼠上述观察指标均显著降低。与模型组比较,鼻渊舒组、新安鼻渊方高剂量组大鼠血中溶菌酶含量显著升高(P<0.05,或P<0.01)。模型组大鼠上颌窦黏膜表面有大量炎性细胞聚积,黏膜上皮变性、脱落,纤毛缺失、排列紊乱,黏膜内及黏膜下大量炎性细胞浸润,病理分级显著增高,鼻渊舒组,左氧氟沙星组,新安鼻渊方高、中剂量组病理分级显著降低。结论新安鼻渊方对大鼠急性鼻窦炎有较好的防治作用。
Objective To study the preventive effects of Xin'an Biyuan Fang (XABYF), a traditional Chi- nese prescription for sinusitis, against acute sinusitis in rats. Methods Of 90 SD rats, 10 were randomized into control group, while other 80 were used to induce acute sinusitis modei. According to the scoring standard of sinusitis symptoms, 60 rats with more than 6 scores were averagely randomized into model group, high (15.2 g/kg)-, middle (7.6 g/kg)-, and low-dose (3.8 g/kg) XABYF treated groups, Biyuan- shu (a patent Chinese medicine for sinusitis, 3.5 ml/kg) treated group, and levofloxacin (40 mg/kg) trea- ted group. Rats were treated with intragastric injection of corresponding drugs seven days after model in- duced. Rats in the normal group and the model group were treated with the same volume of normal saline. The period of administration was seven days. The scores of sinusitis symptoms, body temperature, white blood cells (WBC) counts, neutrophil (N) percentage, serum contents of lysozyme, and histopathological changes of paranasal sinuses were observed. Results The scores of sinusitis symptoms, body tempera- ture, WBC counts, and N percentage in the model group were significantly higher than those in the normalcontrol group. Indicators mentioned above in levofloxacin treated group, Biyuanshu treated group, and high-, middle- and low-dose XABYF treated groups were significantly decreased. Compared with those in the model group, serum contents of lysozyme in Biyuanshu treated group, high-dose XABYF treated group were significantly increased. In the model group, a large number of inflammatory cells aggregated on the surface of mucous membrane of maxillary sinus. Mucosal epithelium degenerated and desquamated. Cilia were absent and arranged disorderly. A great quantity of inflammatory cells within and under the mucous mem- brane infiltrated. The pathological grade in the model group were increased significantly, while that in Biyuanshu treated group, levofloxacin treated group, and high- and middle-dose XABYF treated groups were decreased significantly. Conclusion XABYF has good protective effects against acute sinusitis in rats.
