超高效液相色谱法测定生栀子和焦栀子药材中栀子苷含量 被引量:6

Content Determination of Geniposide in Fructus Gardeniae and Baked Fructus Gardeniae Using Ultra Performance Liquid Chromatography

摘要 目的采用超高效液相色谱法(ultra performance liquid chromatography,UPLC)建立栀子药材中栀子苷的含量测定方法。方法采用BEH C18色谱柱(1.7μm,2.1mm×100mm),流动相为乙腈-水(梯度洗脱),检测波长238nm,流速为0.3ml/min,柱温35℃。结果栀子苷进样量在0.015~0.240μg范围内,与峰面积线性关系良好(y=0.056 2 x-0.075 6,r=0.999 1),生栀子的平均回收率为98.98%,相对标准偏差(relative standard deviation,RSD)为0.40%(n=5);焦栀子的平均回收率为98.12%,RSD=0.47%(n=5)。结论超高效液相色谱法简便、快速、准确、重复性好,可用于栀子药材中栀子苷的检测。 Objective To establish an ultra performance liquid chromatography (UPLC) method for determining the contents of Geniposide in the medical materials of Fructus Gardeniae. Methods BEH C18 chromatographic column (1.7μm, 2. 1 minX 100 mm) was employed, with acetonitrile-water (gradient elution) being mobile phase, wavelength being set at 238 nm, flow rate being 0.3 ml/min, and column temperature being 25 ℃. Results Within the range of 0. 015 to 0. 240 μg, Geniposide had a good linear relation to the peak area (y=0.056 2 x-0.075 6, r=0. 999 1). The average recovery rate of Geniposide was 98.98% in Fructus Gardeniae (relative standard deviation was 0.40%, n=5) and 98.12% in baked Fructus Gardeniae(relative standard deviation was 0.47%, n= 5). Conclusion UPLC can be used for determining Geniposide in medical materials of Fructus Gardeniae Because of its simplicity, rapidness, accuracy, and perfect reproducibility.
