摘要 用90%饱和度的硫酸铵盐析沉淀纤维素酶蛋白,将蛋白沉淀用3%戊二醛交联固定制得交联酶聚集体;研究交联酶聚集体的最适反应温度、最适反应pH、酸碱稳定性、热稳定性、批次稳定性。交联酶纤维素酶聚集体的最高比活性为559.2 U/g湿载体,最适反应温度为60℃,最适pH为9.0。交联酶聚集体的热稳定性和pH稳定性均优于游离酶,重复使用10批活性基本稳定。交联纤维素酶聚集体稳定性较好,具有一定的工业应用前景。 Cellulase was precipitated by ammonium sulfate and then cross-linked with glutaraldehyde,as a result,cross-linking cellulase aggregation was obtained.Finally,the optimum temperature,optimum pH,thermal stability,pH stability and batch cycle numbers of immobilized enzyme were studied.The highest enzyme activity obtained was 559.2 U/g wet carriers.The optimum pH of the immobilized enzyme was 9.0,and the optimum temperature was 60°C,almost no activity lost after 10 cycles.Furthermore,cross-linking cellulase aggregation had better thermal and pH stability than free enzyme.Cross-linking cellulase aggregation had good operating stability,which had the potential for industrial biocatalyst use.
