摘要
用薄层色谱法对处方中的黄连、栀子、五味子、羌活进行定性鉴别,用高效液相色谱法测定制剂中黄芩苷的含量,色谱条件为:色谱柱为Diamonsil C18(4.6 mm×200 mm,5μm);流动相为甲醇体积∶水体积∶冰醋酸体积(50∶50∶1);流速为1 mL.min-1;柱温为35℃;检测波长为274 nm。薄层色谱能检出黄连、栀子、五味子、羌活的特征斑点,黄芩苷在0.354~3.54μg范围内呈良好线性关系,r=0.999 8,平均回收率为99.77%,RSD值为0.32%,5批样品黄芩苷含量平均值为3.92 mg.g-1。结论:本试验建立的方法专属性强,可作为黄栀护肝片的定性定量检测方法。
To establish the method to distinguish Huangzhi hugan tablet by TLC and determine the content of baicalin by HPLC,coptis chinensis Franch,Gardenia jasminoides Ellis,Schisandra chinensis(Turcz.)Baill,Notopterygium incisum Ting ex H.T.Chang were identified by TLC.The content of baicalin was determined by HPLC.The samples were separated on Diamonsil C18 HPLC column with the mobile phase of methanol-water-glacial acetic acid(50∶50∶1).The flow-rate was 1.0 mL·min-1 with the colume temperature of 35 ℃.The detection wavelength was 274nm.Feature spots of Coptis chinensis Franch,Gardenia jasminoides Ellis,Schisandra chinensis(Turcz.)Baill.Notopterygium incisum Ting ex H.T.Chang were detected in the thin layer chromatography(TLC).The linear relationship of Baicalin was well between 0.354-3.54 μg(r=0.999 8),the average recovery was 99.77%,RSD was 0.32%.The average content of baicalin in five batches of Huangzhi Hugan Tablet was 3.92 mg·g-1.The specificity of above experiments was well.The methods can be used to distinguish Huangzhi Hugan Tablet and determinate the content of baicalin.
