猪圆环病毒Ⅱ型和伪狂犬病弱毒疫苗体外共接种对猪外周血单个核细胞调节性细胞因子mRNA表达的影响 被引量:4

摘要 为分析猪圆环病毒Ⅱ型(PCV2)对伪狂犬病弱毒苗(PRV)免疫反应的影响。用Real-time PCR技术检测了仔猪外周血单个核细胞(PBMC)体外单接种和共接种PCV2与PRV后调节性细胞因子IL-4、IL-10、IL-12p40及IFN-γ的mRNA表达水平。结果表明:PRV能引起IL-4、IL-12p40与IFN-γ的mRNA表达上调,PCV2能引起IL-4、IL-10与IL-12p40的mRNA表达上调;而且,PCV2能抑制PRV诱导IL-4、IL-12p40及IFN-γ的mRNA表达上调,其中对IFN-γmRNA表达的抑制效果最为显著(P<0.05),提示PCV2可能会对PRV的细胞免疫反应产生不利影响。 To explore the effect of porcine circovirus type Ⅱ(PCV2) on the immune response of attenuated pseudorabies vaccine(PRV),peripheral blood mononuclear cells(PBMCs) of a piglet were experimentally inoculated with PCV2 and PRV alone or in combination,respectively.The mRNA level of regulatory cytokines including IL-4,IL-10,IL-12 p40 and IFN-γ in piglet PBMCs at different hours post inoculation were quantitatively measured by real-time PCR method.The results showed that there was up-regulation of the mRNA expression of IL-4,IL-12 p40 and IFN-γ induced by PRV,IL-4,IL-10 and IL-12 p40 induced by PCV2,respectively.Moreover,the up-regulation of IL-4,IL-12 p40 and IFN-γ mRNA expression induced by PRV was repressed in presence of PCV2,and the repression effect of IFN-γ mRNA expression was significant(P0.05).The results revealed that PCV2 could induced the detrimental effect on the cell-mediated immune response of PRV.
