rAd-p53联合伊立替康诱导结肠癌细胞株HT-29凋亡的实验研究 被引量:3

摘要 目的观察重组人p53腺病毒(rAd-p53)注射液、伊立替康(CPT-11)两药单用和联用对结肠癌细胞株HT-29体外增殖、细胞凋亡的影响及可能机制。方法取对数生长期HT-29细胞随机分为rAd-p53组、CPT-11组、联合组及对照组,其中rAd-p53组分别加入感染剂量(MOI)为50~800的rAd-p53注射液,CPT-11组分别加入质量浓度为10.0~0.1μg/mL的CPT-11;联合组同时加入上述剂量的rAd-p53注射液和CPT-11,筛选协同作用最佳组为400 MOI rAd-p53注射液+5μg/mL CPT-11;空白组及对照组加入等体积RPMI1640培养基。采用MTT法检测细胞抑制率,流式细胞仪测定细胞周期和凋亡率,Western blot检测Livin蛋白表达,实时荧光定量PCR检测LivinmRNA表达。结果 rAd-p53组、CPT-11组及联合组细胞增殖均受到抑制,细胞凋亡率及G2/M期分布比例均显著高于对照组,Livin mRNA及蛋白表达均显著低于对照组,尤以联合组为著(P<0.05、0.01),且呈时间—剂量依赖方式。结论 rAd-p53和CPT-11联用可能通过下调Livin mRNA及蛋白表达协同抑制HT-29细胞增殖并诱导其凋亡;此为结肠癌的基因治疗提供了新思路。
