摘要
目的:研究依达拉奉对糖尿病周围神经病变(DPN)大鼠坐骨神经多聚ADP核糖聚合酶(PARP)表达及细胞凋亡的影响。方法:SD大鼠80只,10只作为正常组;其余70只采用链脲佐菌素(STZ)一次性腹腔注射诱导建立DPN模型,随机抽取40只,并随机分为对照组和治疗组,各20只;治疗组每天给予依达拉奉(3mg/kg)腹腔注射,共4周;观察各组大鼠坐骨神经PARP表达和细胞凋亡指数(AI)。结果:与正常组相比,对照组大鼠坐骨神经PARP表达及细胞AI明显增高(P<0.01),治疗组PARP表达及细胞AI较对照组明显降低(P<0.01)。结论:依达拉奉可降低DPN大鼠坐骨神经损伤,其机制可能与降低PARP表达及细胞凋亡有关。
Objective: To investigate the effects of edaravone on expression of poly(ADP-ribose) polymerase(PARP) and cell apoptosis on the sciatic nerve in rats with experimental diabetic peripheral neuropathy.Methods: Among 80 health male SD rats,10 were assigned to normal group,and the diabetic peripheral neuropathy model was generated on the remaining 70 rats by a single intraperitoneal injection of streptozotocin.Out of the 70 rats,40 were randomly divided into the control(n=20) and treatment(n=20) groups.Rats in the treatment group have received edaravone(3 mg/kg) intraperitoneal injection daily for consecutive 4 weeks.The expression of PARP and cell apoptosis index(AI) in the sciatic nerve were evaluated and compared.Results: When compared with the rats in the normal group,the expression of PARP and AI in the sciatic nerve significantly increased in the control rats(P0.01).However,the expression of PARP and AI were significantly decreased in treatment group(P0.01) relative to those in the control rats.Conclusion: Edaravone treatment helps to lower the damage occurred in the sciatic nerve in rats with experimental diabetic peripheral neuropathy,perhaps by way of attenuating oxidative damage,down-regulating PARP expression,and inhibiting cell apoptosis.
