摘要
建立了液相色谱-串联质谱法同时测定Beagle犬血浆中的右旋布洛芬和左西替利嗪。使用C18色谱柱,以乙腈-甲醇-5 mmol/L乙酸铵溶液(25︰56.25︰18.75)为流动相,采用电喷雾电离源、多反应监测模式,正负离子切换扫描。右旋布洛芬和左西替利嗪在0.2~100μg/ml和0.008~4μg/ml范围内线性关系良好,日内和日间RSD小于8.7%,提取回收率大于80.9%。
A LC-MS/MS method was established for the determination of dexibuprofen and levocetirizine in Beagle dog plasma. A C18 column was used with the mobile phase of acetonitrile-methanol-5 mmol/L ammonium acetate solution (25 : 56.25 : 18.75). A tandem mass spectrometric detection was conducted using multiple reaction monitoring (MRM) under the positive/negative ionization switching mode. The calibration curves of dexibuprofen and levocetirizine were linear in the ranges of 0.2 - 100 μg/ml and 0.008 - 4 μg/ml. The intra- and inter-day RSDs were less than 8.7% and the extraction recoveries were over 80.9 %.
