摘要
目的探讨microRNA(miRNA,miR)在重型乙型肝炎(SHB)患者外周血单个核细胞(PBMC)中的表达及意义。方法用RNA-DNA嵌合探针液态芯片(flexible multiple analyte profiling,xMap)技术检测40例慢性乙型肝炎(CHB)患者、30例SHB患者和40例体检健康对照者(NC)PBMC中miR-191、-223、-222、-145、-21、-31、-126、-20a、-372的表达水平。结果 CHB组、SHB组、NC组PBMC中miR-191、-222、-145、-31、-372表达无统计学差异(P均>0.05);而miR-21、-126和-223有统计学差异(F分别为64.607、86.412和54.648,P均<0.05)且均在SHB组表达最低[(1.417±0.580),(2.801±0.631),(6.658±0.545)]。3组间miR-20a表达有统计学差异(F=69.163,P<0.05),SHB组miR-20a表达最高(0.586±0.065)。结论 SHB组miR-21、-126、-223表达下调和miR-20a表达上调可能参与SHB的发生、发展。
Objective To explore the expression profile of miRNA in peripheral blood mononuclear cells(PBMC) of severe hepatitis B(SHB) patients.Methods The RNA-DNA probe liquid chip(flexible multiple analyte profiling,xMap) technique was used to detect the expression levels of miR-191,-223,-222,-145,-21,-31,-126,-20a and-372 in the PBMC taken from 40 cases of chronic hepatitis B patients(CHB),30 cases of SHB patients and 40 cases of normal health controls(NC).Results No statistical difference of expression levels of miR-191,-222,-145,-31 and-372 in PBMC were shown among the three groups(P0.05),but the expression levels of miR-21,miR-126 and miR-223 showed statistically significant differences among the three groups(F=64.607,P0.05;F=86.412,P0.05 and F=54.648,P0.05).The lowest expression level existed in SHB group,i.e.,(1.417±0.580) for miR-21,(2.801±0.631) for miR-126 and(6.658±0.545) for miR-223.There was statistically significant difference for expression of miR-20a among the three groups(F=69.163,P0.05),and the highest expression(0.586±0.065) existed in SHB group.Conclusion The downregulation of expression of miR-21,-126,-223 and the upregulation of miR-20a expression may associate with SHB which provide a new clue for the early diagnosis and monitoring of SHB.
