低功率射频联合长春瑞滨长循环热敏脂质体抗肿瘤疗效的实验研究 被引量:2

Antitumor effect of combination low-power radiofrequency with long-circulating temperature sensitive vinorelbine liposomes on VX2 tumor in rabbits

摘要 目的探讨低功率射频联合长春瑞滨长循环热敏脂质体对兔VX2肿瘤的抑瘤效果。方法应用超声引导下组织块包埋法建立新西兰大白兔股部肌肉内VX2肿瘤模型。待肿瘤生长至0.8~1.0 cm时,根据肿瘤大小按区组随机的方法将24只荷瘤兔分成4组。对照组(CON):静脉注射等量生理盐水;单纯射频组(RF):给予1 W 30 min射频治疗;长春瑞滨长循环热敏脂质体组(LCTSL):给予1.8 mg/kg长春瑞滨长循环热敏脂质体;长春瑞滨长循环热敏脂质体联合射频组(LCTSL+RF):射频作用(1 W 30 min)前给予1.8 mg/kg长春瑞滨长循环热敏脂质体。治疗分别在第1、4、7天,共3次。治疗前及治疗后超声连续观测肿瘤的大小,治疗结束后8 d,处死实验兔测量肿瘤重量,计算抑瘤率,标本进行病理检查。结果所有实验兔均能耐受治疗完成实验。LCTSL+RF组抑瘤率为87.2%,显著高于RF和LCTSL组,差异有统计学意义(P<0.05);单纯射频作用无明显抑瘤效果。病理检查显示单纯射频作用无明显肿瘤坏死,而长春瑞滨长循环热敏脂质体联合射频可见明显的肿瘤细胞损伤及坏死。结论低功率射频联合长春瑞滨热长循环热敏脂质体对兔VX2肿瘤有明显的抑瘤效果;射频消融联合长春瑞滨长循环热敏脂质体可能会增大消融范围,提高抗肿瘤疗效。 Objective To investigate antitumor effect of combination radiofrequency with long-circulating temperature sensitive vinorebine liposomes on VX2 tumor in rabbits.Methods VX2 muscle tumor models were established in 24 New Zealand white rabbits through sonographically guided percutaneous puncture tumor tissue implantation.Animals were randomly assigned to 4 groups,and treated with saline,RF alone,temperature-sensitive vinorebine liposomes alone and temperature-sensitive vinorebine liposomes with RF,respectively.Treatments were performed for 3 times at days 1,4,7.Tumor volume was evaluated using ultrasonography prior-treatment and every other days posttreatment.Tumor inhibition rate were calculated.Animals were sacrificed at day 8 after last treatment,and tumors were observed photomicrographically.Results Most of the rabbits well tolerated a vinorebine dose of 1.8 mg/kg.Significantly higher tumor inhibition rate was demonstrated in rabbits treated with combination of RF and long-circulating temperature-sensitive vinorebine liposomes,as compared with the rabbits treated with RF or long-circulating temperature-sensitive vinorebine liposomes alone(P0.05).Photomicroscopic examination revealed obvious tumor necrosis in the tumor treated with combination of RF and temperature-sensitive vinorebine liposomes.Conclusions Low-power RF combined with temperature-sensitive vinorebine liposomes can results definite antitumor effect in rabbits VX2 tumor models,and this primary result may have clinical implication in increasing tissue coagulation if temperature-sensitive vinorebine liposomes combined with RF ablation in clinical treatment.
