屋尘螨抗原致敏大鼠支气管上皮细胞白介素6、白介素8和转化生长因子β1的表达 被引量:1

摘要 目的探讨屋尘螨抗原(Derpl)对大鼠原代支气管上皮细胞白介素6(IL-6)、IL-8和转化生长因子β1(TGF—β1)表达的影响。方法将实验随机分成正常对照组和实验组,实验组中细胞分别暴露于3个不同的Derpl浓度(1、5、10mg,/L)及3个不同的培养时间点(4、8和24h)。然后用逆转录-聚合酶链式反应(RT—PCR)及酶联免疫吸附试验(ELISA)分别检测细胞及细胞上清液中IL-6、IL-8和TGF-β1的表达及含量。结果与对照组相比,实验组各指标有明显变化。Derpl刺激4h,IL-6和IL-8表达开始升高,直至8~24h呈高水平。而在Derpl浓度≥5mg/L条件下,IL-6和IL-8表达最明显,与正常对照组相比差异有统计学意义(P〈0.05)。而TGF—β1则随着时间和浓度的增加,其表达水平呈逐渐上升趋势,各时间点的表达差异均有统计学意义(P〈O.05)。各炎症指标(IL-6,IL-8,TGF-β1)ELISA结果与PCR趋势基本一致。结论Derpl刺激气道上皮细胞,引起支气管上皮细胞免疫反应,释放一系列炎症因子如IL-6、IL-8和TGF-β1,从而参与哮喘气道炎症反应及哮喘的气道重塑。 Objective To investigate the effects of exposure to increasing concentration of Derpl on the expressions of IL-6,IL-8,TGF-β1 in primary rat bronchial epithelial ceils. Methods Ceils were divided into two groups, control groups and experimental groups. In the experimental groups ceils were cultured with 3 different concentrations of derpl (1,5,10 mg/L) for 3 different time (4,8,24 h), then detected mRNA expression and cytokine release by using RT-PCR and ELISA. Results There was a obvious change in the level of cytokine production compared with the control groups. The levels of IL-6 and IL-8 began to ris'e at 4 h, and reach to high expression at the 8-24 h. The maximum concentrations were detected Cytokine release was different. IL 6, IL-8 was at concentrations of ≥5 mg/L ( P〈0.05). But the release of TGF-β1 was Positively related to exposure time and Derpl concentration ( P 〈0.05). The release of IL-6,IL 8 and TGF-β1 level as well as an increase in the expression of IL 6,IL-8 and TGF-β1 mRNA manifested evidently. Conclusions The Derpl allergen can stimulate the release of inflammatory cytokines from the rat bronehia epithelial ceils, such as (IL-6, IL-8, and TGF-β ). It is suggested that HDM-induced cytokines may play an airway inflammation and remodeling of allergic asthma.
