摘要
目的测定无机活性元素(德莫林)对大鼠皮肤慢性溃疡组织细胞增殖的影响,并探讨其作用机制。方法制备SD大鼠慢性溃疡模型,实验组应用无机活性元素,对照组常规消毒换药。于用药第3、7天时取2组溃疡局部肉芽组织,应用流式细胞术检测细胞周期;RT-PCR技术对用药7d时2组肉芽组织增殖相关基因PCNA、CyclinD1、P27的表达进行检测。结果与对照组比较第3、7天时实验组G1期细胞比例明显低于对照组(P<0.05),而S期细胞比例则高于对照组(P<0.05);第7天时实验组CNA、Cy-clinD1mRNA表达均高于对照组(P<0.05),而P27mRNA表达低于对照组(P<0.05)。结论无机活性元素有促进慢性溃疡肉芽组织增殖的作用,其机制与该药物能促进PCNA、CyclinD1、P27表达有关。
Objective To investigate the effects of inorganic active elements (Dermlin TM) on the cell proliferation of chronic ulcer tissue in animal model, and to explore its action mechanism. Methods The animal models with chronic ulcers were established in SD rats. The rats in experimental group were treated with inorganic active elements, however, the rats in control group were treated by routine disinfection dressing. The cell cycle was detected by flow cytometry on 3 days and 7 days after administration. The expressions of PCNA, CyclinD1, P27 were measured on 7 days after treatment. Results As compared with that in control group, the cell ratio at G1 stage in experimental group on 3 days and 7 days after treatment was significantly lower than that in control group ( P 〈 0.05), however, the ratio at S stage was significantly higher than that of control group ( P 〈 0. 05 ). The expressions of CAN, CyclinD1 mRNA in experimental group on 7 days were significantly higher than those of control group, however, the expression of P27 was obviously lower than that of control group ( P 〈 0. 05 ). Conclusion The inorganic active element ( Dermlin TM) has the effect of promoting proliferation of chronic ulcer granulation tissue,and its action mechanism is related with its effect of up-regulating the expressions of PCNA, CyclinD1 and P27.
