摘要 目的:探讨SEMA3B基因在不同病理类型来源肺癌细胞株中的表达及意义。方法:采用实时定量逆转录聚合酶链反应(Real Time RT-PCR)和琼脂糖凝胶电泳方法,检测来源于不同病理类型肺癌细胞株中SEMA3B基因的表达水平。结果:在不同病理类型的细胞株中SEMA3B-mRNA均较正常肺癌组织显著低表达;在大细胞肺癌细胞株H 1 299、腺癌细胞株PC7、鳞癌细胞株LK2或LC/Sq1细胞中SEMA3B均表达缺失。结论:SEMA3B低表达或表达缺失可能是肺癌发生的机制之一,深入分析SEMA3B在肺癌细胞中与其它信号通路间的相互作用,将有利于我们阐明肺癌的发生。 Objective:To study SEMA3B expression in human lung cancer cell lines with different pathological classification and its significance.Methods:We performed semi-quantification reverse transcription-polymerase chain reaction(RTPCR) and agarose gel electrophoresis method to tested SEMA3B expression in hunman lung cancer cell lines.Results:SEMA3B mRNA was lower in lung cancer cell lines compared with normal human lung tissue,and didn't express in large-cell neuroendocrine carcinoma H1299 cell line and adenocarcinoma cell line PC-7 and squamous cell carcinoma cell line LK2 or LC/Sq1 by 2% agarose gel electrophoresis.Conclusion:Low or absent SEMA3B gene expression in lung cancer cell lines may be one of mechanism in leading to lung cancer.Further study for the interrelationship between SEMA3B signaling with other signaling pathways will help us to clarify the occurrence of lung cancer.
