MC-LR对小鼠卵巢颗粒细胞活性及雌孕激素分泌的影响

摘要 目的研究微囊藻毒素LR(MC-LR)对小鼠卵巢颗粒细胞活性及雌孕激素分泌的影响。方法所制备21日龄雌性小鼠卵巢颗粒细胞,以0、5、10和20μg/mL MC-LR染毒,分别于24、48和72h检测其增殖活性及雌孕激素分泌情况。结果染毒24h,随着剂量增加其增殖活性上升,20μg/mL剂量组高于对照组;染毒48和72h,其增殖活性下降,20μg/mL剂量组低于对照组。染毒24h,各剂量组的雌二醇分泌量增加,均高于与对照组,随着染毒时间延长分泌量下降;至72h,各剂量组的分泌量均明显低于对照组。随着染毒剂量增加和时间延长,孕酮分泌有不同程度的上升。结论 MC-LR对小鼠卵巢颗粒细胞具有细胞毒性并影响雌二醇和孕酮的分泌。 Objective To study the effects of MC-LR on viability,estradiol and progestonal secretion function of ovarian granulosa cells in mouse. Methods The cells of female mousse were isolated and exposed to 0,5, t0 and 20ug/mL MC- LR respectively. The cell viability,the levels of estradiol and progestone were detected after 24,48and 72 h respective- ly. Results After 24 h, cell viability was increasing with the doses, 20 ug/mL group was higher than that of control group. After 48 and 72 h,cell viability was inhibited,and 20 ug/ml dose group lower than that of control group. After 24 h,the estradiol secretion of the exposure group increased in all dose groups, and was higher than that of control group. With prolonging of exposure time,the secretion of estradiol decreased. As to 72 h, the estradiol secretion of all exposure groups was lower than that of control group significantly. With dose increasing and the exposure time prolon- ging,progestone secretion was increasing to a certain degree. Conclusion MC-LR can induce toxic effects on ovarian granulosa cells and influence the secretion of stradiol and progestone.
