摘要
目的 采用四氮唑盐MTT法研究肿瘤细胞状态对检测壁虎活性单体BH1258209半数抑制浓度IC50的影响.方法 取处于对数生长期的人肝癌HepG2细胞接种于96孔板,细胞贴壁后,分别加入含10%胎牛血清及无胎牛血清的RPMI-1640培养液配制的不同浓度的BH1258209,培养48h,72h后呈色、比色,检测肿瘤细胞生长抑制率,计算IC50.结果 含10%胎牛血清培养液配制的BH1258209(A组)作用的肿瘤细胞处于倍增状态,能使细胞变小、变圆、胞浆浓缩,且抑瘤率较高,HepG2的IC50为0.33g/L.无胎牛血清培养液配制的BH1258209(B组)作用的肿瘤细胞处于非倍增状态,细胞形态亦发生改变,但不如A组明显,其抑瘤率较低,HepG2的IC50为1.38g/L.结论 BH1258209作用于倍增状态的肿瘤细胞的IC50明显低于非倍增状态的IC50,且随着BH1258209浓度的增加,肿瘤细胞的抑制作用更明显.
Objective To study the efleel of tumor cell stale o11 detecting the half inhibitory concentration of gecko active monomer BH1258209 by MTT assay. Methods Human hepatoma HepG2 cells were cultured conventionally. The tumor cells of logarithmic' phase were seeded in 96-well plates. Cells were treated with different concentrations of BH1258209 after adhesion. BH1258209 was prepared with containing 10% fetal serum and fetal bovine sermn-free RPMI-1640 culture medimn. The cells were performed coloration and colorimetry after cultt, red 48h and 72h. The tumor cell growth inhibition rate and caleulat IC50 were detetqed. Results The tumor cells treated with BH1258209 which prepared with culture medium containing 10% fetal bovine serum were in the doubled state,and the ceils became smaller, munded,c-ytoplasm condensed. At the same time the inhibition rate was higher and the IC50 of HepG2 was 0.33g/L. Conversely the tumor cells treated with BHI258209 which prepared without culture medium fetal hovine serum-free were in the non-doubled state, the cells had changed,but not as good as cells in group A. And the inhibition rate was lower,the IC50 of HepG2 was 1.38g/L. Conclusion The IC50, of BH1258209 on tumor cells in doubled state is significantly lower than in non-doubled state. The inhibitor effect on tumor cells is more obviously with the increased concentration of BH1258209.
