摘要
[目的]采用反相高效液相色谱法测定颈复康颗粒中葛根素的含量。[方法]色谱柱为Kromasil 100-5C18(250 mm×4.6 mm,5μm);流动相为无水甲醇-水-磷酸(20∶80∶0.2,V/V/V),流速为1.0 ml/min;检测波长为250 nm。[结果]阴性对照无干扰;计算回归方程为Y=4 233 801X-34 587,R为0.999 9(n=6)。结果表明,在0.063~1.254μg/ml浓度范围内,葛根素浓度与峰面积呈良好的线性关系,R=0.999 9;加样回收率为101.8%,RSD为1.09%(n=9)。[结论]该方法简便、准确、专属性强,可用于颈复康颗粒中葛根素的含量测定。
[Objective] The aim was to determine the content of puerarin in neck-recover-granule using RP-HPLC.[Methods] The chromatographic column was Kromasil 100-5C18(250 mm×4.6 mm,5 μm);mobile phase was absolute methanol-water-phosphoric acid(20∶ 80∶ 0.2,V/V/V),flow velocity was 1.0 ml/min;the detection wavelength was 250 nm.[Result] The negative control showed no interference,the calculated regression equation was Y=4 233 801X-34 587,R=0.999 9(n=6).The results showed that within the concentration of 0.063-1.254 μg/ml,there was good linear relation between puerarin concentration and peak area,R=0.999 9;the recovery rate was 101.8% and RSD was 1.09%(n=9).[Conclusion] The method is simple,convenient and accurate,has strong specificity,can be used to determine the content of puerarin in neck-recover-granule.
