摘要
[目的]检测Pb2+对小鼠淋巴细胞DNA的损伤作用。[方法]应用单细胞凝胶电泳技术(SCGE),研究在不同Pb2+暴露浓度(0.05、0.50、5.00 mg/L)下12 h后对小鼠外周血淋巴细胞核DNA的损伤作用。采用DNA拖尾率、DNA损伤专用单位、DNA损伤级别等指标,探讨细胞DNA损伤与Pb2+处理浓度的相关性。[结果]随着Pb2+浓度的增高,拖尾率、DNA损伤专用单位都呈上升趋势。[结论]重金属Pb2+对小鼠淋巴细胞DNA损伤呈正相关剂量效应关系,应用单细胞凝胶电泳技术可对重金属导致的遗传毒性进行检测。
[Objective]The aim was to detect Pb2+-induced DNA damage in mice's peripherall blood lymphocytes.[Methods] The DNA damage in mice's peripherall blood lymphocytes under different exposed concentrations of Pb2+(0.05,0.50,5.00 mg/L) after 12 h were studied using SCGE,and the correlation between DNA damage and Pb2+ treatment concentration was discussed using DNA trailing rate,DNA damage specialized units and DNA damage grades.[Result] With the increasing of Pb2+ concentration,trailing rate and DNA damage specialized units showed rising trend.[Conclusion] There was a significant dose effect relationship between the heavy metals Pb2+ treatment and DNA damage in mice's lymphocytes,moreover,the genetic toxicity generated by heavy metals can be detected using SCGE.
