摘要
[目的]检测多胺处理对小鼠子宫内膜上皮的影响。[方法]低浓度的腐胺、亚精胺、精胺处理小鼠子宫内膜上皮细胞,利用实时定量PCR检测多胺处理后多胺代谢相关酶类mRNA水平的变化,利用细胞计数、MTT法、流式细胞分析检测细胞增殖能力。[结果]腐胺处理上皮细胞可造成SAMDC、SMO、SMS、ODC、PAO mRNA水平显著下降;亚精胺处理上皮细胞可造成SAMDC、SRM、ODC mRNA水平显著下降,SSAT mRNA水平显著上升;精胺处理上皮细胞可造成SAMDC、SRM、ODC mRNA水平显著下降,SMO、PAO、SSAT mRNA水平显著上升。腐胺和亚精胺可促进子宫内膜上皮细胞的生长和活力增加,但精胺可显著降低细胞生长和活力。[结论]多胺处理可引起子宫内膜上皮细胞内多胺相关酶类mRNA水平的变化,对细胞增殖能力有重要的调节作用。
[Objective] The aim of this study was to investigate the effect of polyamine treatment on mouse endometrial epithelial cells.[Method] After the mouse endometrial epithelial cells were treated with low levels of putrescine,spermidine and spermine respectively,the transcription of polyamine-related genes was detected by real-time PCR,and cell proliferation was disclosed by cell counting,MTT and flow cytometry analysis.[Result] Putrescine obviously decreased the mRNA levels of S-adenosylmethionine decarboxylase(SAMDC),spermine oxidase(SMO),spermine synthase(SMS),ornithine decarboxylase(ODC) and polyamine oxidase(PAO)genes.Spermidine obviously decreased the mRNA levels of SAMDC,spermidine synthase(SRM) and ODC genes and obviously increased the mRNA levels of spermidine/spermine N1-acetyltransferase(SSAT) genes.Spermine obviously decreased the mRNA levels of SAMDC,SRM and ODC genes and obviously increased the mRNA levels of SMO,PAO and SSAT genes.The epithelial cells viability was significantly increased after treatment with putrescine and spermidine,but it was decreased by spermine treatment.[Conclusion] Regulation of polyamine-related genes by polyamine could lead to the changes of polyamine-related gene mRNAs,which might regulate the epithelial cell proliferation.
