摘要
目的用LC-MS/MS法测定大鼠血浆中利福布丁的浓度。方法以替米沙坦为内标,加入0.2 mL血浆,用1 mL乙酸乙酯萃取,高速离心后上清液氮气吹干并用流动相复溶,取20μL进样检测。结果利福布丁的最低检测限为0.5 ng.mL-1,在0.3~9.6μg.mL-1内线性良好,低、中、高浓度的提取回收率均大于85%,批内、批间RSD低于10%,成功检测了大鼠血浆中利福布丁的血药浓度。结论所用方法灵敏度好、准确度高、分析时间短、样品处理更简便,适用于测定利福布丁的血药浓度。
OBJECTIVE To develop an LC-MS/MS method for determination of Rifabutin in rats plasma.METHODS 0.2 mL plasma samples were mixed with 1 mL ethyl acetate followed by extraction and centrifugation.The supernatant was dried by N2 and the re-dissolied by the mobile phase,20 μL of which was detected by LC-MS/MS.RESULTS The method was linear for Rifabutin in the range of 0.3-9.6 μg·mL-1.The LLOD was 0.5 ng·mL-1.The extraction recoveries of low,middle and high concentration were all higher than 85%.The intra-day and inter-day precisions(RSD) were both less than 10%.CONCLUSION The LC-MS/MS method was simple,sensitive and accurate.It was suitable for the determination of rifabutin in plasma.
