摘要
于 1999年 5~ 6月对栉孔扇贝精子进行超低温保存研究。结果表明 ,抗冻液为 DMSO5% +蔗糖 5%并以 2 0℃ / min的降温速率通过 0~ - 80℃时的保存效果最好 ,冻精的活力、受精率、孵化率分别为 4 9.39%、4 2 .0 6 %和 12 .15%。样品体积会对超低温保存效果产生影响 ,体积为 0 .6 ml或 1ml比较适宜。此外 ,在 0℃进行预处理平衡是必须的 ,但时间不宜太长。冻精用 35℃水浴解冻效果最佳 ,50℃次之 ,2 0℃最差。对精子产生冻伤的温域在 - 30~ - 6 0℃之间 ,必须以适当降温速率越过该范围 。
The experiments of cryopreservation of the spermatozoa of Chlamys(Azumapecten) farreri was studied in 1999. Motility, fertility and hatchery rate of 49.39%, 42.06% and 12.15% were obtained in scallops using a cryoprotectants dilution(DMSO5%+sucrose5%) with freezing rate 20℃/min. The volume of samples between 0.6 or 1ml is suitable for cryopreserving spermatozoa of Chlamys(Azumapecten) farreri. Moreover, it is necessary to equilibrate the pre process at 0℃, and 2 minutes is enough for that. The optimal thawing temperature is 35℃ in water bath, 20℃ worst. The freezing damage to the sperm of chlamys (Azumapecten)farreri occurred between -30℃and -60℃, so it is important to choose the optimum freezing rate to cross over it, no halting during the processing.
