摘要
目的建立了高效液相色谱法测定人血清中卡马西平的药物浓度。方法以symmetryshield RP18柱(5.0μm,3.9×150mm)为色谱柱。流动相为甲醇∶水为55∶45,流速1.0mL.min-1。Waters 2487双波长紫外检测器,检测波长210nm。结果线性范围为(2.0~20.0)μg.mL-1,r=0.999852,最低检测限0.15μg.mL-1;平均回收率为96%,高、中、低浓度样品的日间和日内测定相对标准偏差RSD均小于5%。结论本法适用于卡马西平的血药浓度测定。
OBJECTIVE To establish an HPLC method for determining serum level of Carbamazepine.METHODS A symmetryshield RP18 column(5μm,3.9×150mm)was used.The mobile phase,methanol-phosphate solution(55∶45) was used at a flow rate 1.0mL·min-1,The detector is Waters 2487 Dual λ Absorbance Detector,detector was set at the wavelength of 210nm.RESULTS The calibration curve was linear in the range of 2.0 to 20.0μg·mL-1.The average recovery was 96%,and the RSD within-day and between-day were all less than 5%.CONCLUSION This method is suitable for therapeutic drug monitoring of Carbamazepine.
