摘要
小麦成熟胚愈伤组织的诱导率、生长率及胚性愈伤组织发生率的基因型效应有显著差别。供试的 5个基因型中 ,愈伤组织诱导率基因型间差异达到极显著水平。诱导率大小顺序为 :辽 10>774 2 >Roblin>7757>新克旱 9。以成熟胚为外植体的胚性愈伤组织发生率 ,辽 10为 78% ,774 2为 10 % ,新克旱 9为 0 %。外植体与基因型共同作用影响胚性愈伤组织发生率 ,774 2两种外植体(成熟胚与幼穗 ) ,都有胚性愈伤组织发生。但不同外植体发生率不同 ,成熟胚为 10 % ,幼穗为 1%。而新克旱 9两种外植体均无胚性愈伤组织发生 ,并有褐化愈伤组织 (褐化率分别为 6 9.50 %和53.70 % )。
The effect of genotype on the rate of induction,growth and embryogenic callus initiating were different significantly.Callus induction rate showed Liao 10>7742>Roblin>7757>Xin Kehan9.The difference in callus induction rate between genotypes was also significant statistically.The embryogenic callus initiating rate came from the mature embryo explant were Liao 10.80%,7742 around 10%,as to Xin Kehan 9.00% was found.Explant and genotype collation affected on embryogenic callus generation rate.As to explant(mature embryo and immature spike),7742 genotype both generated embryogenic callus,but different explants caused different generating rate,10% in mature embryo and 1% only in immature spike.Xin kehan 9 did not generate embryogenic callus in all explants and caused brow phenomenon(69.5% and 53.70% respectively).Different embryogenic callus generating rates were found on different media.Auxin played an important role in regulating embryogenic callus initiation.$$$$
