摘要
以昆明树木园云南红豆杉 (Taxusyunnanensis)人工林为研究对象 ,采用垂直板式聚丙烯酰胺凝胶电泳 ,检查了此人工林雌配子体的苹果酸脱氢酶 (MDH)、酯酶 (EST)、莽草酸脱氢酶 (SKD)、谷氨酸草酰乙酸转氨酶(GOT)及过氧化物酶 (PER)的同工酶 ,分析了此群体等位酶的遗传变异。根据多态位点的标准 ,发现检测的基因位点的 70 %是多态的。每个基因位点发现的等位基因平均数是 2 10。所检测的 10个位点平均期望杂合度是0 2 91;平均实际杂合度为 0 2 93;平均等位基因有效数是 1 5 98。
Vertical slab polyacrylamide gel electrophoresis was used to examine inheritance of five isoenzymes (MDH,EST,SKD,GOT,PER) in megagametophytes of Taxus yunnanensis artificial forest growing in Kunming Arboretum. Inheritance and differentiation of isoenzymes were analyzed at 10 loci.According to the standard for loci polymorphism,it was found percentage of loci polymorphism was 70%.The average number of alleles detected per locus was 2 10.The average expected heterozygosity of all samples was 0 291,the average observed heterozygosity of all samples was 0 293, and the effective number of alleles per locus was 1 598.The results show gene diversity of the artificial forest is relatively high.
