葡萄籽多酚大孔树脂纯化过程的近红外光谱快速检测 被引量:11

摘要 采用近红外光谱透射法,对葡萄籽提取液大孔树脂层析过程多酚含量进行快速无损测定。以Folin-Ciocalteus比色法为对照方法,运用偏最小二乘法(PLS)建立定量校正模型,测定大孔树脂洗脱过程葡萄籽多酚浓度变化。该模型相关系数R2达到0.9995,校正集误差均方根(RMSEC)和预测集误差均方根(RM-SEP)分别为0.713和2.67,校正集相对偏差(RSEC)和预测集相对偏差(RSEP)分别为2.39%和8.48%。本方法快速、有效、无损,可用于葡萄籽多酚柱层析纯化过程检测和质量控制。 A method was developed for the determination of purifying process of grape seed extracts by near infrared spectroscopy (NIRS) coupled with partial least squares (PLS) algorithm. The colourimetric assay of Folin-Ciocalteus was used as the reference method to determine the content of polyphenol. The samples in the purification process, which did not participate in modeling, were used to test the performance of the models. PLS factors, correlation coefficient of calibration set (R2) , t root mean square of cross-validation (RMSECV), root mean square of prediction (RMSEP), relative standard error of calibration (RSEC) and relative standard error of prediction (RSEP) were used to evaluate the performance of the models. The optimal calibration model was obtained with R2 = 0.9995, RMSEC=0. 713, RMSEP=2. 67, RSEC=2. 39% and RSEP=8. 48% in the 5500- 6200 cm-1 spectral region by using Savitzky-Golay and second derivative spectral transform method. The results showed that the method is fast, nondestructive, environmental friendly, and can be used to determine the active components in maeroporous adsorption resin purification process of traditional Chinese medicine.
