参龙健脑胶囊对脑缺血大鼠脑组织谷氨酸及N-甲基-D-天冬氨酸受体表达的影响 被引量:4

摘要 目的观察参龙健脑胶囊对脑缺血损伤大鼠脑组织谷氨酸(Glu)、N-甲基D-天冬氨酸受体(NMDAR)表达的影响,探讨其神经保护机制。方法结扎双侧颈总动脉制备缺血性脑损伤大鼠模型,模型成功后随机分为假手术组、模型组、脑复康组及参龙健脑胶囊低、高剂量组,参龙健脑胶囊低、高剂量组和脑复康组分别按照0.40、1.20、0.45 g/kg剂量给药,每日灌胃1次,连续28 d。采用比色法观察Glu在大鼠脑组织中的含量,免疫组化方法观察NMDAR亚单位NR1在海马区的表达。结果与模型组大鼠比较,参龙健脑胶囊组大鼠脑组织中Glu含量明显降低(P<0.01,P<0.05),海马区NR1表达减少(P<0.01)。结论参龙健脑胶囊对缺血性脑损伤的保护作用可能与其抑制脑缺血后脑组织中Glu的释放、减少NR1的表达密切相关。 Objective To explore the effects of Shenlong Jiannao capsule on the expression of glutamate(Glu) and N-methyl-D-aspartate receptor 1(NR1) in brain tissue of cerebral ischemia rats.Methods Two-vessel occlusion were adopted to reproduce the cerebral ischaemia animal model.Rats were randomly divided into 5 groups as follows:sham operation group,model group,positive control group with pisacetam,Shenlong Jiannao capsule groups(low and high dosage).Shenlong Jiannao(low and high dosage) capsule groups and pisacetam groups were treated by gavage at the dosage of 0.40,1.20,0.45 g/(kg?d).After 28 days,colorimetry was used to determine the contents of Glu in brain tissue,and immunohistochemistry was used to assay the expression of NR1 in hippocampus of cerebral ischaemia rats.Results The contents of Glu were significantly reduced in brain tissue(P0.01,P0.05),and the expression of NR1 in hippocampus decreased in Shenlong Jiannao capsule group(P0.01).Conclusion Shenlong Jiannao capsule can ameliorate the cerebral injury by restraining the release of Glu and reducing the expression of NR1.
