摘要
从牦牛皮下采集皮蝇(Hypoderma)幼虫,用DNA提取试剂盒、Chelex-100两种方法提取幼虫的基因组DNA;通过PCR扩增线粒体细胞色素氧化酶Ⅰ基因(COⅠ)中的高度可变序列,扩增产物经BfaⅠ酶切后用琼脂糖凝胶检测,结果表明所采集的皮蝇幼虫属于同一种。测序结果显示,扩增的序列与GenBank中中华皮蝇(H.sinense)相应序列的相似性为99.56%,表明所采集的皮蝇幼虫均为中华皮蝇。
RFLP and sequencing was used to identify the Hypoderma larvae collected from yaks.DNA of larvae was extracted by Genomic DNA Extraction Kit and Chelex-100 method.The variable region of the mitochondrial cytochrome oxidase Ⅰ gene(COⅠ) was amplified by PCR.BfaⅠ digestion and agarose gel electrophoresis revealed that the collected larvae belonged to one species.Sequencing and alignment showed that the amplified sequence shared 99.56% homology with that of H.sinense in GenBank,suggesting that the collected larvae were H.sinense.
