不同时间缺氧对脂肪细胞炎症因子表达的影响 被引量:2

Effect of different incubation time of hypoxia on the levels of inflammatory cytokines in adipocytes

摘要 目的:探讨不同时间缺氧孵育后脂肪细胞炎症因子TNF-α和MCP-1表达的变化。方法:常氧(21%O2)和缺氧(1%O2)4、8、16 h孵育脂肪细胞,免疫印迹法检测葡萄糖转运子1(GLUT1)的表达,Real-time PCR法测定脂肪细胞TNF-α和MCP-1基因表达。结果:与常氧组相比,缺氧孵育的脂肪细胞GLUT1蛋白和TNF-α、MCP-1基因的表达均升高(P=0.002、P=0.016),并且随着缺氧时间的延长而显著升高。结论:缺氧造成脂肪细胞GLUT1和TNF-α、MCP-1的表达升高,缺氧时间越长,升高越明显。 Objective: To explore the effect of different incubation time of hypoxia on expression of inflammatory cytokines TNF-α and MCP-1 in adipocytes.Methods: 3T3-L1 adipocytes were treated with normoxia(21% O 2) or hypoxia(1% O 2) conditions for 4,8,or 16 hours.The glucose transporter 1(GLUT1) was detected by Western blot.The mRNA expressions of TNF-α and MCP-1 by adipocytes were determined by real-time PCR.Results: The protein expression of GLUT1 and the mRNA expressions of TNF-α and MCP-1 were higher in hypoxia-treated 3T3-L1 adipocytes than nomoxia-treated cells(P=0.002,P=0.016).Moreover,the expression levels were increased with prolonged incubation time of hypoxia.Conclusion: The mRNA expressions of TNF-α and MCP-1,as well as protein expression of GLUT1 by adipocyte are increased under hypoxia treatment,and the longer incubation time of hypoxia,the more obviously level increases.
