rhEPO对大鼠肠道缺血再灌注损伤的保护作用

摘要 目的:探讨人类重组促红细胞生成素(rhEPO)对大鼠肠道缺血再灌注损伤的保护作用,观察其对肠道紧密连接蛋白(ZO-1)表达的影响。方法:健康雄性Wistar大鼠36只,随机分为假手术组(A组)、缺血再灌注组(B组)和rhEPO预处理组(C组)3组,每组12只。A组只给予剖腹手术,C组缺血前10 min预先皮下注射rhEPO 1 000 U/kg,B、C组均夹闭肠系膜前动脉45 min后恢复再灌注2 h,取血清及小肠组织行二胺氧化酶(DAO)、D-乳酸、TNF-α检测及HE染色和ZO-1蛋白免疫组织化学检测。结果:与B组比较,A、C组小肠绒毛排列整齐,破坏程度明显减轻,ZO-1蛋白表达水平增高。ELISA结果显示:血浆DAO、D-乳酸和TNF-α水平在B组和A、C组之间均有显著性差异(P<0.05),A和C组之间差异也有统计学意义(P<0.05)。结论:预先注射rhEPO对缺血再灌注损伤的肠道有保护作用,并且其对ZO-1蛋白的表达有一定影响。 Objective: To explore the protective effect of rhEPO on intestinal ischemia reperfusion in rats and observe its effects on the expression of tight junction protein(ZO-1).Methods: Thirty-six male Wistar rats were randomly divided into three groups,the sham operation group A,the ischemic/reperfusion group B,the rhEPO pretreatment group C,n =12 respectively.Laparotomy was operated in group A,rhEPO 1 000 U/kg were injected subcutaneously 10 minutes before ischemia in group C,superior mesentery artery(SAM) were occlused for 45 minutes ischemia in group B and C,then followed reperfusion 2 hours.Serum diamine oxidase(DAO),D-lactate and TNFα were measured and intestine histopathology was observed by HE staining and the expression of ZO-1 protein were detected by immunohistochemistry.Results: Compared to group B,the arrangement of intestinal villus was better,the damage degree was insignificantly and the expression of ZO-1 protein was higher in group A and C.The serum concentration of DAO,D-lactate and TNF-α in three groups had difference significantly(P0.05),and there was statistical significance(P0.05) in group A and C.Conclusion: RhEPO protects the intestine from ischemia reperfusion injury in rats and have effects on the expression of ZO-1 protein.
