摘要
以草莓品种红颊的叶片为受体,通过对影响遗传转化的一系列因子:潮霉素选择压、抗生素种类和浓度、农杆菌浓度、侵染时间和共培养时间的优化,获得了较高转化效率的体系。结果表明:菌液浓度为OD600=0.4,侵染时间为6 min,选用250 mg/L的Cb作为抑菌抗生素、5 mg/L的潮霉素作为筛选抗生素,共培养时间3 d是一个高效的转化体系,抗性芽诱导率达到25%。
Taking the leaves of strawberry cultivar 'Benihope' as experimental acceptor,a higher-efficient genetic transformation system was obtained through the optimization of series of influential factors,such as hygromycin selection stress,type and concentration of antibiotics,agrobacterial concentration,infection time and co-culture time.The results showed that when the agrobacterial concentration was OD600=0.4,the infection time was 6 min,250 mg/L Cb was used as the antibacterial antibiotics,5 mg/L hygromycin was used as the screening antibiotics,and the co-culture time was 3 days,this transformation system was highly efficient,and its inducement rate of resistant shoots reached 25%.
