金蒿抗流感滴丸对小鼠TNF-α、IL-2和IFN-γ等细胞因子的影响

Effect of Jinhao Anti-Flu Pills on Cytokines of TNF-α,IL-2 and IFN-γ in Mice

为了观察金蒿抗流感滴丸对小鼠细胞因子的影响,对其灌胃给药7d后采血制备血清,采用ELISA法测定血清中肿瘤坏死因子α(TNF-α)的含量;以ConA诱导小鼠脾淋巴细胞增殖,ELISA法测定细胞上清液中白细胞介素2(IL-2)和γ干扰素(IFN-γ)的含量。结果显示:金蒿抗流感滴丸0.05、0.1和0.2(g/kg)/d剂量组及利巴韦林组和复方一枝蒿颗粒组均能显著促进小鼠血清中TNF-α的分泌(P<0.05);金蒿抗流感滴丸浓度为78.13～5000μg/mL,对ConA诱导的小鼠淋巴细胞分泌IFN-γ和IL-2均有一定的促进作用。结论:金蒿抗流感滴丸对TNF-α、IL-2、,IFN-γ等细胞因子的分泌具有一定的调节作用,这可能是其发挥抗流感病毒作用的免疫机理之一。

The objective of this study was to observe the effect of Jinhao anti-flu pills on the cytokines of mice.Tumor necrosis factor α（TNF-α） in the serum was measured by ELISA method after serum preparation using the blood sampled from the mice which were gavaged for seven days.Spleen lymphocytes were amplified under the ConA induction.Interleucocyte 2（IL-2） and γ Interferon（IFN-γ） in the supernatant fluid were measured by the ELISA method.The results showed that dosage groups of 0.05,0.1 and 0.2（g/kg）/d of Jinhao anti-flu pills,Ribavirin group,and compound Artemisiae rupestris granules group all significantly promoted the secretion of TNF-α in the mouse serum（P0.05）.Jinhao anti-flu pills in the concentration range of 78.13～5000 μg/mL could promote ConA-induced secretion of IFN-γ and IL-2 from the lymphocytes of mice.Conclusion：Jinhao anti-flu pills have a certain regulating effect on the secretion of such cytokines as TNF-α,IFN-γ,IL-2,which may be one of the immunity mechanisms of its anti-influenza function.
