丙氨酰-谷氨酰胺二肽对大鼠创伤性脑损伤后应激性肝损害的干预作用

Protection Effect of Alanyl Glutamine on Hepatic Stress Injury Induced by Traumatic Brain Injury in Rats

摘要 【目的】探讨丙氨酰-谷氨酰胺二肽(Ala-Gln)对创伤性脑损伤(TBI)大鼠应激性肝损害(HSI)的干预作用。【方法】将60只健康雄性Wister大鼠随机分为假手术对照组(A组)、TBl组(B组),和Ala-Gin干预组(C组)。C组再分四个亚组分别于致伤后不同时间点3~24h给予Gln干预,各组大鼠均于伤后48h处死取材,检测血清丙氨酸转氨酶(ALT)和天门冬氨酸转氨酶(AST),测定肝组织超氧化物歧化酶(SOD)和丙二醛(MDA)水平,观察肝脏组织病理学改变。【结果】B组的ALT、AST较A组明显升高,SOD值明显低于A组,MDA显著高于A组,肝脏组织病理学可见变性、坏死等改变;c组各时点ALT、AST及MDA均较B组显著下降,SOD值明显升高,肝脏组织病理学改变显著改善,且伤后早期干预效果明显。【结论】TBI后可出现肝损伤,应用Ala-Gln对TBI后的肝损伤具有干预作用,且此保护作用可能通过改善肝组织的氧化应激状态完成,并具有显著的时效相关性。 [Objective] To explore the interventional effect of alanyl glutamine(Ala-Gln) on hepatic stress inju- ry induced(HSI) by traumatic brain injury(TBI) in rats. [Methods]Sixty healthy male Wister rats were randomly divided into sham operation group(control group, group A), TBI group(group 13) and Ala-Gln intervention group (group C). Group C was given Gin intervention at different time points after injury. At 48h after injury, the rats in each group were sacrificed for detecting the levels of alanine transaminase(ALT) and aspartate aminotransferase (AST) in serum, superoxide dismutase(SOD) and malondialdehyde(MDA) in liver tissue. The pathological chan- ges in liver were observed. [Results] Compared with the control group, the levels of ALT and AST in TBI group significantly increased. SOD in TBI group was obviously lower than that in control group, while MDA in TBI group was obviously higher than that in control group. The pathology of liver showed the changes such as degener- ation and necrosis, etc. After Ala-Gln intervention, ALT, AST and MDA significantly decreased in comparison with group B, while SOD level obviously increased, and the pathological changes in liver were improved significantly. Early intervention after injury, the effects were even more obvious. [Conclusion] TBI can induce HSI. Ala-Gln has interventional effect on hepatic injury after TBI. The effect may be completed by improving oxidative stress state in liver and has obvious time-action relationship.
