摘要
目的研究脓毒症大鼠血浆NPY水平变化与肠黏膜屏障损害相关性。方法 60只SD大鼠随机分成假手术组(n=30),脓毒症组(n=30),假手术组仅行盲肠探查术,脓毒症组采用CLP制作大鼠脓毒症模型。2组术后即刻皮下注射生理盐水5ml。各组均于术后3、6、12、24、48 h时点,采集腹主动脉血液标本测NPY、DAO。结果血浆中NPY水平均高于正常值,在CLP术后3 h较正常对照组显著升高(P<0.05),达最高值,以后逐渐降低,至48 h组仍高于正常组(P<0.05);血浆二胺氧化酶(DAO)水平在CLP术后3 h开始升高,以后逐渐升高趋势,至48 h显著高于正常(P<0.05)。NPY与DAO经过相关分析呈高度负相关(r=-0.974,P<0.01)。结论脓毒症致急性肠道损伤后血浆中的NPY早期呈现高浓度状态,DAO亦高于正常水平,NPY升高可能是造成肠缺血缺氧的因素之一。
Objective The study of the correlation between levels of serum neuropeptide Y and the damage of intestinal barrier in septic rats.Methods Sixty SD rats were divided into two groups randomly: sham group,n=30 and sepsis group,n=30.Sham group underwent exploratory surgery of the cecum only,and sepsis group underwent cecal ligation and puncture(CLP) to induce rat model of sepsis.In each group,5ml saline was subcutaneously injected at once.Blood samples in abdominal aorta were collected for each group at 3h,6h,12h,24h,48 h time point to measure the levels of NPY and DAO.Results Plasma NPY levels were higher than normal,3h after CLP they were significantly higher compared with normal control group(P0.05) and reached the highest value,then gradually rose and reduced,48 h group was still higher than the normal group(P0.05);plasma diamine oxidase(DAO) levels began to increase at 3 h after CLP,then showed gradual increasing tendency to 48 h,which was significantly higher than normal(P0.05).NPY and DAO showed a high negative correlation through correlation analysis(r =-0.974,P0.01).Conclusion Plasma NPY after sepsis-induced acute intestinal injury shows high concentrations in the early state,DAO is also higher than normal levels.The increasing serum NPY levels maybe result in intestinal ischemia and hypoxia.
